Samantha Ruth Prabhu was born on April 28, 1987, in Alappuzha, Kerala, to Joseph Prabhu, a Telugu father, and Ninette Prabhu, a Malayali mother. She was raised as the family’s youngest child in Pallavaram, Tamil Nadu, with two elder brothers: Jonathan Prabhu and David Prabhu.
In 2012, Samantha featured in live action Telugu-Tamil bilingual project Eega (Naan Ee). The film received positive reviews in both Telugu and Tamil languages and it was one of the highest-grossing Telugu films of the year, earning ₹1.15 billion globally. Her performance as a micro artist who runs a NGO and takes revenge for the murder of her lover earned her the Filmfare Award for Best Telugu Actress.
Despite her cross upbringing, she identifies as a Tamilian. She received her education at Holy Angels Anglo Indian Higher Secondary School and then went to Stella Maris College in Chennai to earn a degree in commerce.

Samantha Ruth Prabhu debuted in the Telugu film titled Ye Maaya Chesave, directed by Gautham Vasudev Menon (2010).
